结缕草属植物杂交后代形态特征和SRAP分子标记鉴定

Morphological Characteristics and Identification of SRAP Molecular Markers in Hybrid Progeny of Zoysia

摘要: 本研究通过形态特征和相关序列扩增多态性(Sequence-related amplified polymorphism,SRAP)分子标记技术对结缕草属植物种间杂交获得的19个组合34个后代的形态变异和杂种真实性进行了鉴定。结果表明:同一组合不同后代在10个外部性状上均存在不同程度的差异;方差分析结果发现大部分杂交后代的外部性状与母本或双亲存在显著性差异;根据形态特征34个后代中有30个后代初步被鉴定为真杂种;SRAP分子标记鉴定结果显示,34个杂交后代均具有父本的特异性DNA片段,全部被鉴定为真杂种。本研究中的结缕草属植物杂种真实性鉴定结果为杂交后代的进一步研究及应用提供了科学依据。

Abstract: The authenticity of 34 hybrid progenies from 19 interspecific hybridized combinations was identified by morphological characteristics and SRAP molecular markers. The results showed that variations of 10 morphological variables existed among different progenies in one hybrid combination. The variance analysis results showed that the morphological characteristics of most progenies were significantly different from their female parent or both parents. 30 of 34 progenies were identified to be true hybrids according the morphological characteristics. The identified results with SRAP molecular markers showed that all of the 34 progenies had the special DNA fragments of male parent and were identified as true hybrids. Authenticity identification results of Zoysia hybrids in this study could provide scientific evidence for the further research and application of hybrids.
