青海草原毛虫化学感受蛋白基因的鉴定与组织表达分析

Identification and Expression Profiling of Chemosensory Protein Genes in Gynaephora qinghaiensis(Lepidoptera:Lymantriidae)

摘要: 青海草原毛虫(Gynaephora qinghaiensis)是青藏高原上严重危害高寒草甸的重大害虫。本研究基于青海草原毛虫转录组测序数据,利用生物信息学方法及qRT-PCR技术,对GqinCSPs基因进行鉴定及表达谱分析,共鉴定出11个GqinCSPs基因,均具有完整的编码区序列。其中,GqinCSP5GqinCSP8两个基因没有信号肽,GqinCSP4GqinCSP9相似性最高,二三级结构主要由α螺旋和无规则卷曲组成,系统进化表明青海草原毛虫CSPs与棉铃虫和柑橘凤蝶的亲缘关系较近。基因表达谱分析显示,11个青海草原毛虫CSP基因在雌雄成虫头部、雄虫触角以及2龄至7龄阶段中均有表达,且体现不同的表达水平。GqinCSP1GqinCSP3GqinCSP5GqinCSP8GqinCSP9在2龄阶段的相对表达量显著高于其他阶段(P<0.05)。本研究为深入了解青海草原毛虫化学感受机制奠定基础,同时也为寻找青海草原毛虫绿色防治的分子靶标提供理论依据。

Abstract: Gynaephora qinghaiensis is a serious pest of the alpine meadow on the Qinghai-Tibet Plateau. In this study,based on the transcriptome sequencing data of G. qinghaiensis,the identification and expression profile of GqinCSPs genes were performed using bioinformatics methods and qRT-PCR,11 GqinCSPs genes were identified,all of them containing complete coding region sequences. GqinCSP5 and GqinCSP8 had no signal peptide, GqinCSP4 had the highest similarity to GqinCSP9,and the secondary and tertiary structures were mainly composed of α helix and random coil. The phylogeny suggested that CSPs of G. qinghaiensis were closely related to those of Heliothis armigera and Papilio xuthus. Gene expression profiling showed that 11 CSP genes were expressed in the heads of male and female adults,male antennae and the second and seventh instar stages with different expression levels. The relative expression of GqinCSP1,GqinCSP3,GqinCSP5,GqinCSP8,and GqinCSP9 was significantly higher in the second instar than in the other stages (P<0.05). This study lays a foundation for deep understanding the chemosensory mechanism in G. qinghaiensis,and provides a theoretical basis for finding the molecular targets of green prevention and control.
