狗牙根诱变后代生长速度评价及DNA指纹图谱构建

Evaluation of Growth Rate and DNA Fingerprint Construction of Mutated Progenies from Bermudagrass

摘要: 狗牙根(Cynodon dactylon)是一种常见的暖季型草,利用诱变技术处理现有品种进行改良,是快速选育优良新品种的一种途径。本研究以国审品种‘阳江’狗牙根通过60Co-γ辐射处理获得的12个诱变后代为材料,进行生长速度评价和基于序列相关的扩增多态性(SRAP)分子标记技术的DNA指纹图谱构建。结果表明,诱变后代间匍匐茎总长度、地上部分干重、匍匐茎数量、地下部分干重等4个指标均存在显著的变异。对这4个指标进行隶属函数分析,对诱变后代的生长速度进行综合评价,认为其生长速度由快到慢依次为M37>M16>M1>M25>M10>M18>M28>M29>M26>‘阳江’>M22>M31>M4。利用12对SRAP引物对12个诱变后代和6个狗牙根主栽品种进行指纹图谱构建,其中7对引物组合可以直接对18份参试材料进行准确鉴定。本研究为进一步国产狗牙根新品种的选育和保护提供了可靠的实验依据。

Abstract: Bermudagrass (Cynodon dactylon) is a prevalent warm-season grass. Mutation technology offers a rapid method for selecting new superior varieties. This study used 12 mutated progenies from the nationally approved ‘Yangjiang’ bermudagrass variety,obtained through 60Co-γ radiation mutagenesis treatment. The growth rate was evaluated,and a DNA fingerprint was constructed using sequence-related amplified polymorphism (SRAP) molecular marker technology. The results showed that there were significant variations in the total stolon length,aboveground dry weight,stolon number,and underground dry weight among the mutated progenies. Membership function analysis was performed on these four indicators to comprehensively evaluate the growth rate of the mutated progenies. The growth rate,from fastest to slowest,is M37>M16>M1>M25>M10>M18>M28>M29>M26>‘Yangjiang’>M22>M31>M4. Twelve pairs of SRAP primers were used to construct fingerprint maps of 12 mutated progenies and 6 main bermudagrass varieties. It was found that 7 pairs of primer combinations could directly and accurately identify 18 test materials. This study provides a reliable experimental basis for the breeding and protection of new domestic bermudagrass varieties.
