草地早熟禾成熟胚离体培养植株再生技术的研究

A Study on the Technique of Plant Regeneration from Mature Seed Embryo of Kentucky bluegrass in vitro

摘要: 以大青山草地早熟禾和超级伊克利早熟禾的成熟种子为外植体材料,研究激动素和生长素及其不同配比组成的激素组合对愈伤组织诱导发生与生长状态及其绿苗分化能力的影响。结果表明,在愈伤组织诱导培养基中附加2.0mg/L2,4-D和0.2mg/LBA,诱导频率在30%-43%之间。在其继代培养基中添加1.0mg/L2,4-D和0.5mg/LBA,愈伤组织增殖速度快,且保持胚胎发生能力。在愈伤组织分化培养基中附加2.0mg/LBA或CPPU,经继代培养3代的愈伤组织的绿苗分化率保持在30%左右。在继代培养中,早代选择外表呈干燥、紧密、颗粒状愈伤组织是克服随着继代培养时间的延长愈伤组织再生植株的能力显著下降这一障碍的简易有效方法。

Abstract: Mature seeds of Poa pratensis L.cv.Daqingshan and Total eclipse were used for explants.Effect of hormone compositions of different kinds and ration on callus induction,growth state and shoot regeneration was studied.Results indicated that the frequency of callus induction reached 30%-43% in the callus induction medium supplemented with 2.0 mg/L 2,4-D and 0.2 mg/L BA.During subcultures,rapid growth of callus and embryogenesis were maintained in the callus subculture medium plus 1.0 mg/L 2,4-D and 0.5 mg/L BA.After callus subcultures,the rate of shoot regeneration from callus was about 30% in the differentiation medium supplemented with 2.0 mg/L BA or CPPU.Selecting the callus of dry,compact,small pellet was a simple effective method in early generation to overcome the obstruction of plant regeneration.
