人工老化对纳罗克非洲狗尾草种子活力、生理生化指标的影响

摘要:

为探明非洲狗尾草(Setaria sphacelata ‘Narok’)种子贮藏过程中生理生化变化及种子活力下降的主要原因,以收获后未贮藏的非洲狗尾草种子为材料,进行人工加速老化处理,测定其发芽率、种子活力及生理生化指标的变化。结果表明,短期人工加速老化有助于打破纳罗克非洲狗尾草种子休眠,提高种子活力,而长时间老化处理会使种子活力下降。其中,老化1~2 d的种子活力最高,为对照的1.52~1.72倍(P<0.05);人工老化处理能显著提高纳罗克非洲狗尾草种子保护酶活性(P<0.05),且随老化时间的延长,酶活性基本呈先升高后降低的趋势;老化处理后,纳罗克非洲狗尾草种子可溶性糖和丙二醛含量显著增加(P<0.05),可溶性蛋白含量随老化时间的延长表现为先升高后降低再升高的趋势(P<0.05),萌发抑制物活性呈先降低后增加的趋势(P<0.05)。分析认为,在人工加速种子老化过程中,可能同时存在休眠解除与老化劣变两种作用力,它们共同决定了种子活力的变化。

Abstract:

In order to ascertain the physiological and biochemical changes and the main reason for the vigor descent of Setaria sphacelata ‘Narok’ seeds, the seeds of freshly harvested ‘Narok’ were selected and treated by means of artificial accelerated aging, and the changes of their germination rate, seed vigor and the physiological characteristics were investigated. The results showed that a short term artificial accelerated aging treatment helped to break the dormancy of S. sphacelata ‘Narok’ seeds, improve seed vigor, while that in long time could reduce seed vigor. Among them, the seed vigor of age 1~2 d was the highest, which was 1.52 to 1.72 times of the control (P<0.05). Artificial aging treatment could significantly improve protective enzyme activities of S. sphacelata ‘Narok’ seeds (P<0.05) and with the extension of aging time the enzyme activity showed a general trend of increasing first and then decreasing;After aging treatment, the soluble sugar and MDA contents of S. sphacelata ‘Narok’ seeds was increased significantly (P<0.05). Furthermore, with the extension of aging time the soluble protein contents showed a trend of first increasing and then decreasing and then increasing (P<0.05), while the germination inhibitor activity showed a general trend of decreasing first and then increasing (P<0.05);It was believed that during the process of artificial accelerated aging of seeds, there might be two kinds of forces acting on dormancy release and aging deterioration, which together determined the change of seed vigor.
