盐酸脱绒处理对白羊草种子抗氧化性能的影响

Effects of Delinting Treatment with Hydrochloric Acid on Oxidation Resistance of Bothriochloa ischaemum

摘要: 为探讨盐酸(HCl)脱绒处理对白羊草(Bothriochloa ischaemum)种子抗氧化性能的影响,本试验用不同浓度(体积分数为30%,40%,50%,60%和70%)的HCl溶液分别脱绒处理白羊草种子0(CK)min,1 min,5 min,10 min和15 min后,分析其脂质过氧化含量及抗氧化酶活性的变化规律。结果表明:白羊草种子抗氧化性能的变化与HCl浓度及其引发时间密切相关;脱绒处理1~10 min,HCl浓度30%时,白羊草种子内过氧化氢酶(Catalase,CAT)和抗坏血酸过氧化物酶(Ascorbate peroxidas,APX)活性均显著高于CK(P<0.05),过氧化物酶(Peroxidase,POD)活性显著升高(P<0.05),而H2O2和丙二醛(Malondialdehyde,MDA)含量均显著低于CK(P<0.05);而HCl浓度70%时,超氧化物歧化酶(Superoxide dismutase,SOD)活性降低,POD活性升高,CAT活性始终高于CK,此时H2O2含量不断升高。隶属函数综合评价分析表明,利用30%的HCl处理5 min是农业生产中白羊草种子脱绒的最佳方法,本研究为白羊草种子生产提供依据。

Abstract: In order to investigate the effects of hydrochloric acid (HCl) delinting treatment on the oxidation resistance of Bothriochloa ischaemum, seeds were treated with different concentrations (volume fractions of 30%, 40%, 50%, 60% and 70%) of HCl solution for 0 (CK) min, 1 min, 5 min, 10 min and 15 min, respectively, and the changes of lipid peroxidation content and oxidation enzyme activity were analyzed. The results showed that the changes of the oxidation resistance of Bothriochloa ischaemum were closely related to the concentrations of HCl and the times of its priming. When the delinting treatment was lasted for 1 to 10 minutes and the HCl concentration was 30%, the activities of catalase and ascorbate peroxidase in the seeds of Bothriochloa ischaemum were significantly higher than those in the CK (P<0.05), and the activity of peroxidase significantly increased (P<0.05), while the contents of H2O2 and malondialdehyde were significantly lower than those in the CK (P<0.05). However, when the HCl concentration was 70%, the activity of superoxide dismutase decreased, the activity of peroxidase increased, and the activity of catalase was higher than that in CK. Meanwhile, the content of H2O2 continuously increased. The comprehensive evaluation and analysis of membership function showed that the treatment of 30% HCl for 5 min was the best way to delint the seeds of Bothriochloa ischaemum in agricultural production, and this study provided a basis for the seed production of Bothriochloa ischaemum.
