蒙农杂种冰草AcdMYB1基因克隆及表达分析

Cloning and Expression Analysis of the AcdMYB1 Gene from Agropyron critatum × A.desertorum ‘Hycrest-Mengnong’

摘要: 蒙农杂种'冰草(Agropyron cristatum×A. desertorum 'Hycrest-Mengnong')具有干草产量高,适口性好,抗逆性强的特点,是中国北方干旱地区建立人工草地的适宜草种。本研究利用同源克隆方法从'蒙农杂种'冰草中克隆得到的1个MYB类转录因子基因(命名为AcdMYB1),对其进行了生物信息学分析和表达分析。结果表明:该基因cDNA序列长度为1 161 bp,包含完整开放阅读框(ORF)为969 bp,编码322个氨基酸,具有MYB转录因子的保守结构域。AcdMYB1与小麦族的山羊草、普通小麦、野生二粒小麦的同类基因亲缘关系最近,序列一致性均在90%以上。组织表达中AcdMYB1基因在穗中的表达量最高,其次为叶、根,茎中表达量最低;在模拟自然干旱胁迫下,能够显著降低AcdMYB1的表达水平。烟草亚细胞定位显示,该蛋白定位于细胞核。本研究为进一步探索'蒙农杂种'冰草AcdMYB1基因及MYB转录因子家族提供了科学依据。

Abstract: Agropyron cristatum×A. desertorum 'Hycrest-Mengnong' is a pasture variety cultivated by Inner Mongolia Agricultural University and it a tetraploid grass (2n=28) with high hay yield,good palatability and strong resistance to stress. It is a suitable grass species for establishing artificial grassland in arid areas of north China. A MYB transcription factor gene was cloned from A. cristatum×A.desertorum 'Hycrest-Mengnong' by homology-based cloning,named as AcdMYB1,the bioinformatics analysis and expression analysis were carried out on. The results showed that a cDNA sequence with a length of 1 161 bp and a maximum open reading frame of 969 bp encoding 322 amino acids,the AcdMYB1 sequence had a conserved domain of MYB transcription factor. AcdMYB1 had a genetic relationship with Aegilops tauschii,Triticum aestivum and Triticum dicoccoides by more than 90% sequence homology. The expression analysis results showed that the expression of AcdMYB1 gene was the highest in the spike,followed by leaves,roots and stems. Under a drought stress,the expression level of AcdMYB1 was significantly reduced. The protein was localized in the nucleus. The study provided a scientific basis for further research on the AcdMYB1 gene and the MYB transcription factor family in A. cristatum×A.desertorum ‘Hycrest-Mengnong' .
