野生黄花苜蓿胚性愈伤诱导研究

Study on Embryogenic Callus Induction of Wild Medicago falcata L.

摘要: 为优化黄花苜蓿胚性愈伤培养条件,本研究以黄花苜蓿(Medicago falcata L.)为试验材料,探讨培养基、外植体、激素浓度以及凝固剂对黄花苜蓿胚性愈伤形成的影响。结果表明,以成熟胚为外植体接种至MS培养基的愈伤块呈淡黄色,质地为粘性且体积较大,该处理下添加1 mg·L-12,4-D和0.05 mg·L-1KT愈伤诱导率可达95.24%。利用该培养条件进一步探究了凝固剂对愈伤组织诱导的影响,发现培养基中添加6 g·L-1琼脂和7 g·L-1植物凝胶可以有效降低愈伤组织褐化率。因此,诱导黄花苜蓿最适培养基为:以成熟胚为外植体,其成分包括MS培养基,添加1 mg·L-12,4-D+0.05 mg·L-1KT+6 g·L-1琼脂/7 g·L-1植物凝胶。

Abstract: In order to optimize the induction of embryonic callus in alfalfa (Medicago falcata L.) from Xinjiang,the effects of culture medium,explants,hormone concentration and gelling agents on the formation of alfalfa callus tissue were investigated. The results showed that the callus inoculated into MS medium with mature embryo as explants were light yellow,viscous in texture and large in volume,the callus induction rate reached 95.24 % when 1 mg·L-12,4-D and 0.05 mg·L-1KT were added. The effect of coagulant on callus induction was further explored by using this culture condition,it found that the addition of 6 g·L-1 agar and 7 g·L-1 phytagel to the medium effectively reduced the browning rate of callus. Therefore,the optimal medium for inducing alfalfa was mature embryos as explants,and the medium composition was MS+1 mg·L-12,4-D+0.05 mg·L-1KT+6 g·L-1 agar or 7 g·L-1 phytagel.
