青绿苔草(Care breviculmis)愈伤组织诱导与再生体系的建立

摘要: 为建立青绿苔草高效的再生体系,本研究以青绿苔草的种子为外植体,探究不同植物激素配比对愈伤组织诱导、愈伤分化和生根的影响。结果表明:MS+1.5 mg·L-1 NAA+0.5 mg·L-1 6-BA+2.0 mg·L-1 2,4-D为诱导愈伤的最佳培养基,诱导率可以达到68.3%,愈伤表现为松散的黄色颗粒;MS+1.0 mg·L-1 NAA+1.0 mg·L-1 6-BA为最佳分化培养基,分化率为93.8%,不定芽数量达到40个,长势良好;不定芽在1/2 MS培养基中生根效果最好,其根长、株高和生根数量均显著高于其他处理。本研究建立了高效的青绿苔草再生体系,为研究青绿苔草的遗传转化及应用生物技术育种奠定了基础。

Abstract: In order to establish an efficient regeneration system of C. breviculmis,we explored the effects of different plant hormones on the callus induction,differentiation and rooting by using the seed of C. breviculmis as an explant. The results showed that MS+1.5 mg·L-1 NAA+0.5 mg·L-1 6-BA+2 mg·L-1 2,4-D was the best medium for callus inducion,the induction rate reaches 68.3%,and the callus appeared as loose yellow particles. MS+1 mg·L-1 NAA+1 mg·L-1 6-BA was the best differentiation medium with a differentiation rate of 93.8%. There were about 40 adventitious buds growing well. Adventitious buds exhibit the best rooting effect on 1/2MS medium,and their root length,plant height and rooting number were significantly higher than other treatments. In this study,an efficient regeneration system of C. breviculmis was established,which laid a foundation for the study of genetic transformation and application of biotechnology breeding of C. breviculmis.
