热研二号柱花草组织培养研究

Abstract: Studies on tissue culture of Stylosanthes guianensis REYAN Ⅱshow that cotyledon is apromising explant.Callus and shoots can be induced from cotyledon cultured on the medium containingbasic salt of MS, 0.8% agar, 3.0% sucrose, 1.0mg/L NAA and 4.0mg/L BA, pH6.0.Rooting canbe induced from one-month-old shoots cultrued on the mediurn containing half strength MS basic salt,0.8% agar, 1.0% sucrose, 0.5mg/L NAA, and 0.005% active cbarcoal, pH6.0 and the rooting rateis 60%, 25ug/ml, 50ug/ml of Kanamycin inhibits callus formation and redifferentiation respectively.

摘要: 研究结果表明,利用热研二号柱花草无菌苗的子叶为外植体,在M2培养基(MS基本成分+0.8%琼脂+3.0%蔗糖+1.0mg/L萘乙酸+4.0mg/L激动素,pH6.0)培养可诱导分化出愈伤组织和枝条。1月龄枝条在M7培养基(1/2MS基本成分+0.8%琼脂+1.0%蔗糖+0.5mg/L萘乙酸+0.05%活性炭,pH6.0)培养可诱导生根,生根率60%,获得了具有根、茎、叶的完整柱花草组培植株。25和50μg/ml浓度的卡那霉素均能完全抑制子叶形成愈伤组织和愈伤组织再分化。
