大花金挖耳细胞悬浮培养及其胞内代谢产物化感潜力的研究

Abstract: Allelopathic effects of intracellular secondary metabolic products and cell suspension culture of Carpesium macrocephalum Franch.et Sav.are not well reported.In order to optimize cell suspensing culture conditions of C.macrocephalum and to investigate the allelopathy of extracts from C.macrocephalum suspension cells,the effects of different factors on the cell growth and the accumulation of total flavonoids were studied during C.macrocephalum suspension culture under the MS,1/2MS,B5,1/2B5,NT and 1/2NT media,respectively.Results showed that NT medium was suitable for cell cultures according to cell growth and the high content of flavonoid.Effects of five macronutrients on cell growth and flavonoid synthesis were significantly different between treatments(P<0.05).The highest flavonoid content(1.38%) was obtained when the total nitrogen concentration was 50.62 mmol·L-1 and the ratio of NH4+/NO3- in the medium was 2/3.The best concentrations for H2PO4-,Ca2+,Mg2+ and K+ were 5.00 mmol·L-1,1.50 mmol·L-1,0.50~1.00 mmol·L-1,14.39~27.81 mmol·L-1,respectively.Also,ethanol extracts of cell cultivation products from C.macrocephalum had herbicidal activity against 5 tested plant seeds.EC50 values of ethanol extracts to Medicago sativa L.,Cephalanoplos segetum(Bge.) Kitam.,Amaranthus retroflexus L.,Conyza canadensis(L.)Cronq.and Triticum aestivum L.were 10.66,12.32,15.85,102.16,210.96 mg·mL-1,respectively.

摘要: 为了优化大花金挖耳(Carpesium macrocephalum Franch.et Sav.)细胞悬浮培养的条件,评价其胞内代谢产物的化感潜力。本文研究了无机盐浓度不同的MS,1/2MS,B5,1/2B5,NT和1/2NT等6种培养基及NT中N,P,Ca,Mg,K等5种大量元素含量对大花金挖耳悬浮培养细胞生物合成黄酮的影响,并采用微量活性测定法测定了其细胞培养物的化感潜力。结果表明:大花金挖耳细胞悬浮培养的最优基本培养基是NT,NT中总氮浓度为50.62 mmol·L-1,NH4+:NO3-为2:3时,黄酮含量最高为1.38%,H2PO4-,Ca2+,Mg2+,K+等离子依次在5.00,1.50,0.50~1.00,14.39~27.81 mmol·L-1范围内有利于大花金挖耳悬浮培养细胞的生长和黄酮合成;大花金挖耳悬浮培养细胞的粗提物对供试的紫花苜蓿(Medicago sativa L.)、刺儿菜(Cephalanoplos segetum(Bge.)Kitam.)、反枝苋(Amaranthus retroflexus L.)、小白酒草(Conyza canadensis(L.)Cronq.)和小麦(Triticum aestivum L.)等5种植物幼苗根生长均具有抑制作用,毒力依次为10.66,12.32,15.85,102.16,210.96 mg·mL-1
