Fingerprint Constructing and Genetic Diversity Analyzing of Sorghum Sudanense and Sorghum bicolor × Sorghum sudanense with SSR Markers

国审苏丹草和高丹草品种SSR指纹图谱构建及遗传多样性分析

Abstract:

Twenty pairs of SSR primers were screened from 108 pairs to explore the DNA fingerprint constructing and genetic diversity analyzing of 10 Sorghum Sudanense varieties and 7 Sorghum bicolor×Sorghum sudanense varieties. The results showed that 20 pairs of primers totally amplified 121 bands, in which 117 bands were polymorphic and the average value of polymorphic information content (PIC) was 0.703. The number of varieties which could be identified using single pair primer ranged from 2 to 14 and the number of primer mix which could be used to identify 17 varieties ranged from 2 to 5. The genetic diversity analyses of 17 varieties showed that the average Nei's genetic diversity index (H) was 0.480, the average information Shannon diversity index (I) was 0.820, and the similarity coefficients of 17 national varieties ranged from 0.58 to 0.96 as well. These results indicated that the national variety of Sorghum Sudanense and Sorghum bicolor×Sorghum sudanense were rich in genetic diversities.

摘要:

利用SSR分子标记技术,从108对引物中筛选出20对在国审10个苏丹草(Sorghum Sudanense)和7个高丹草(Sorghum bicolor×Sorghum sudanense)品种中进行了指纹图谱构建及遗传多样性分析。结果表明:20对引物在17个品种中共扩增出121条谱带,其中多态性条带有117条,多态性信息含量(PIC)平均为0.703;单对引物能将17个品种区分为2~14个类型,2~5对引物指纹图谱可以将17个品种区分开,可作为鉴别高丹草和苏丹草品种的分子依据;17个品种材料的平均Nei's基因多样性指数(H)为0.480,平均Shannon多样性信息指数(I)为0.820,品种间的相似系数介于0.58~0.96之间,可见,国审苏丹草和高丹草品种具有丰富的遗传多样性。
