Identification and Salt Stress Response Analysis on NAC Gene in Sesuvium portulacastrum

海马齿NAC基因的鉴定及盐胁迫响应

Abstract: NACs belonging to plant specific transcription factor gene family,and play an important role in the response of plant to biotic and abiotic stress. In this study,a total number of 10 NAC family genes were identified based on the transcriptome sequencing data of Sesuvium portulacastrum. Protein structure and physicochemical properties analysis showed that in the 10 SpNACs,the number of amino acids ranged from 164 to 651,the molecular weight from 18400.34 Da to 68678.02 Da,and the isoelectric point from 4.34 to 8.52. All SpNACs were hydrophilic proteins localized in the nucleus;among the 10 SpNACs,only SpNAC02 and SpNAC08 had the transmembrane structures. Conservative motif prediction found out that most of SpNACs contained three motifs,but SpNAC05 contained no motif,SpNAC09 only contained the motif2,and SPNAC07 only contained the motif2 and the motif3. Phylogenetic analysis showed that all SpNACs were clustered into six subfamilies. qRT-PCR was used to analyze the expression of SpNACs under different salt stresses. The results showed that 9 genes did express in root,stem and leaf except for SpNAC03 which did not express in stem and leaf. The expression of 10 SpNACs genes was inhibited in a degree of salinity (40‰ NaCl). These studies enriched the information of NAC gene family members in plants,and provided a reference for screening and identifying the stress-resistant genes of S. portulacastrum.

摘要: NAC基因是仅存于植物中的一类特殊的转录因子,在植物响应生物和非生物胁迫中发挥重要作用。本研究基于转录组测序数据分析,在海马齿(Sesuvium portulacastrum)中共鉴定出10个NAC家族成员,对其蛋白质结构及其理化性质进行分析,结果表明:10个SpNACs的氨基酸数量在164~651之间,分子量大小在18 400.34 Da~68 678.02 Da之间,等电点分布在4.34~8.52之间,均为定位于细胞核的亲水性蛋白,其中SpNAC02和SpNAC08具有跨膜结构。保守基序预测发现多数SpNACs中存在三个motif,但SpNAC05不含motif,SpNAC09只含motif2,SpNAC07只含motif2和motif3。聚类分析结果10个SpNACs聚类在6个亚家族。利用qRT-PCR分析表明在盐胁迫下除SpNAC03在茎、叶不表达以外,其它9个基因均在根、茎和叶中表达。盐生环境下(40‰ NaCl)10个NACs基因的表达均受到不同程度的抑制。以上研究丰富了植物NAC基因家族成员信息,为筛选和鉴定海马齿抗逆基因提供参考。
