燕麦萌发期耐盐性综合评价

Abstract: The purpose of this study was to establish an efficient method for identifying the salt tolerance of oats (Avena sativa L.) during the germination stage, and to screen our excellent parents for salt-tolerant breeding. Using the petri dish paper germination method, 332 oat germplasms were subjected to a salt stress treatment with 0.80% (136.89 mmol·L-1) NaCl solution. Eight indicators such as germination potential were measured. The salt tolerance was comprehensively evaluated and classified by combining the weighted membership function method with cluster analysis. Salt stress significantly inhibited all eight measured indicators. Correlation analysis revealed that highly significant positive correlations among these indicators. The 332 germplasms were classified into five salt tolerance grades: 34 highly salt-tolerant, 83 salt-tolerant, 116 moderately salt-tolerant, 51 salt-sensitive, and 48 highly salt-sensitive accessions. Regression analysis identified the salt tolerance index of root length (STI-RL) and the salt tolerance index of bud fresh weight (STI-BFW) as key indicators for evaluating salt tolerance during germination. Based on two-year evaluations, six materials demonstrated stable salt tolerance and can be utilized as parents in salt-tolerant breeding programs. Comparative analysis indicated that a higher proportion of Avena sativa L. accessions exhibited stronger salt tolerance compared to Avena nuda L. accessions. The 34 highly salt-tolerant germplasms provided excellent resources for salt-tolerant breeding and gene mining.

摘要: 本研究旨在建立高效的燕麦(Avena sativa L.)萌发期耐盐性鉴定方法,并为耐盐育种筛选优良亲本。采用培养皿纸上发芽法,以0.80%(136.89 mmol·L-1)NaCl溶液对332份燕麦种质进行盐胁迫处理,测定发芽势等8项指标,结合加权隶属函数法与聚类分析综合评价其耐盐性并划分等级。盐胁迫对8项指标均产生抑制;相关性分析显示各指标间存在极显著正相关;332份种质被划分为5个等级,包括34份高耐盐、83份耐盐、116份中等耐盐、51份盐敏感及48份高盐敏感种质;通过回归分析确定根长耐盐指数、芽鲜重耐盐指数为萌发期耐盐性评价的关键指标;根据两年评价结果,6份材料的耐盐性稳定,可作为耐盐育种亲本;比较分析显示,耐盐性皮燕麦的比率高于裸燕麦。34份高耐盐种质为耐盐育种与基因挖掘提供了优异资源。
