PEG前处理对饲用燕麦种子EMS诱变效应的影响

Abstract: To optimize the EMS-induced mutagenesis technology of forage oat (Avena sativa L.) and improve mutagenic efficiency, seeds of Oat cultivar ‘Qinghai 444’ and ‘Zhongtian No. 4’ were used as materials in this study. PEG osmotic priming was combined with EMS chemical mutagenesis to screen for optimum compound mutagenesis conditions for forage oat. Based on the germination characteristics and seedling traits of the M1 generation in the two oat cultivars, the pretreatment conditions of 20% PEG for 3 h and 10% PEG for 6 h were found to promote seedling growth. The optimal EMS treatment conditions were 1% EMS for 16 h for ‘Zhongtian No.4’ and 0.75% EMS for 16 h for ‘Qinghai 444’. Through a comprehensive analysis of field agronomic traits at the milk stage in the M2 generation after combined PEG and EMS treatments, it was found that 30% PEG for 3 h combined with the optimal EMS treatment produced the best results for developing germplasm with high-yield potential in both cultivars. Under these conditions, the mutagenesis efficiencies of the M2 population were 2.40% for ‘Zhongtian No. 4’ and 1.26% for ‘Qinghai 444’, respectively, and a total of 22 germplasm lines with high-yield potential were obtained. These results provide a rich material basis for subsequent oat breeding.

摘要: 为优化饲用燕麦(Avena sativa L.)诱变技术,提高饲用燕麦的诱变效率,本研究以‘青海444’和‘中天4号’种子为材料,采用PEG渗透调节前处理协同EMS化学诱变处理,筛选高效的饲用燕麦复合诱变条件。综合两个燕麦品种M1代种子发芽特征及种苗特性,发现20% PEG 3 h和10% PEG 6 h 前处理条件能够促进幼苗生长;两个燕麦品种的最适EMS处理条件分别为1% EMS 16 h(‘中天4号’)和0.75% EMS 16 h(‘青海444’)。通过对PEG和EMS复合处理后M2代乳熟期田间农艺性状综合分析,发现两个品种在30% PEG处理3 h协同最适EMS处理条件,在创制高产潜力种质方面效果最佳,其M2代诱变效率分别为2.40%(‘中天4号’)和1.26%(‘青海444’),共获得具有高产潜力的种质材料22份,为后续燕麦育种提供了变异丰富的材料基础。
