油菜素内酯对低温胁迫下紫花苜蓿生长及生理特性的影响

Abstract: In order to explore the effects of spraying brassinosteroid (BR) on the growth and physiological characteristics of alfalfa (Medicago sativa) seedlings under low temperature stress, the new cold-resistant alfalfa line (P4) and Longdong alfalfa (Medicago sativa ‘Longdong’) were used as the research objects in this study. After 13 days of low temperature stress (4℃), different concentrations of exogenous BR (0.00, 0.25, 0.50, 0.75, and 1.00 mg·L-1, representing T0, T1, T2, T3, T4 treatment, respectively) were sprayed on the leaves. After 5 days, the morphological and physiological indexes of the seedlings were measured. The results showed that low temperature stress significantly inhibited the growth of alfalfa seedlings. However, after applying different concentrations of BR, the plant height, leaf length, leaf width, leaf area, aboveground and underground biomass, root shoot ratio, total root length, total root surface area, root diameter, root volume, root tip number, root activity, phenylalanine ammonia lyase, cinnamic acid-4-hydroxylase, 4-coumaric acid coenzyme A ligase and total flavonoid content of alfalfa seedlings were significantly higher than those of T0P<0.05). The content of malondialdehyde decreased significantly (P<0.05). The comprehensive evaluation of membership function showed that different concentrations of BR treatments were in the order of T2>T1>T3>T4>T0, indicating that spraying 0.25-1.00 mg·L-1 BR could alleviate the effects of low temperature on the growth of alfalfa seedlings, while the 0.5 mg·L-1 (T2) BR treatment was the best. The results provided a theoretical basis for the rational use of BR to solve the problem of chilling injury in alfalfa cultivation.

摘要: 为探明低温胁迫下喷施油菜素内酯(Brassinosteroid,BR)对紫花苜蓿(Medicago sativa)幼苗生长和生理特性的影响,本研究以抗寒苜蓿新品系(P4)和‘陇东苜蓿’(Medicago sativa ‘Longdong)为研究对象,低温胁迫(4℃)13 d后,对叶面喷施不同浓度的外源BR(0.00,0.25,0.50,0.75,1.00 mg·L-1,分别代表T0,T1,T2,T3,T4处理),5天后测定其幼苗形态和生理指标。结果表明:低温胁迫显著抑制了苜蓿幼苗的生长;然而,施加不同浓度BR后,供试苜蓿幼苗株高、叶长、叶宽、叶面积、地上与地下生物量、根冠比、根系总长度、根总表面积、根直径、根体积、根尖数、根系活力、苯丙氨酸解氨酶、肉桂酸-4-羟基化酶、4-香豆酸辅酶A连接酶和总黄酮含量指标均显著高于T0P<0.05);丙二醛含量显著降低(P<0.05)。隶属函数综合评价显示不同浓度BR处理依次表现为:T2>T1>T3>T4>T0,表明喷施0.25~1.00 mg·L-1的BR均能缓解低温对苜蓿幼苗生长的影响,但以0.5 mg·L-1(T2)的BR处理效果最优,其研究结果为合理利用BR解决苜蓿栽培中的冷害问题提供了理论依据。
