Effects of Different Concentrations of Hormones on the Regeneration System of Tall Fescue

摘要:

以‘爱瑞3号’为试材,以无菌种子为外植体,对成熟种子愈伤组织诱导、胚性愈伤组织继代和分化的影响因素进行了研究,构建了高频的高羊茅胚性愈伤组织植株再生体系。结果表明:最佳愈伤诱导培养基配方为MS+5.5 mg·L-1 2,4-D+500 mg·L-1 CH;最佳分化培养基配方为MS+2.0 mg·L-1 6-BA+1.0 mg·L-1 KT;最佳生根培养基配方为1/2MS+0.3 mg·L-1 IBA, 其蔗糖含量为20 g·L-1

Abstract:

The impacts of manifold factors on callus induction from mature seeds, subculture production and embryogenic calli differentiation were studied here. Plant regeneration system of tall fescue (Festuca arundinacea Schreb.) embryogenic calli was established using the sterile seeds of Arid 3 variety as explants. The results showed that the best callus induction medium was MS+5.5 mg·L-1 2,4-D+500 mg·L-1 CH; and MS+2.0 mg·L-1 6-BA+1.0 mg·L-1 KT medium performed the best function in the differentiation of tall fescue embryogenic calli. The best rooting medium was 1/2MS+0.3 mg·L-1IBA, and the content of sucrose was 20 g·L-1.
