柳枝稷TIFY基因家族的鉴定与分析

Identification and Analysis of TIFY Gene Family in Switchgrass

摘要: 为探究TIFY基因家族在柳枝稷(Panicum virgatum)中的生物学功能,本研究运用生物信息学手段对柳枝稷基因组中的TIFY家族成员进行了鉴定,并对其染色体定位、进化与结构特点、启动子区顺式作用元件、时空表达模式等进行了分析,且通过定量实验验证了JAZ亚家族部分成员的激素应答与胁迫响应特性。结果表明:48个柳枝稷TIFY基因分属于ZML(7个)、TIFY(1个)与JAZ(40个)3个亚家族,分布在除染色体3以外的染色体上,且在染色体9成簇排列;柳枝稷TIFY基因按照进化关系及结构特点可分为5组,每组内的成员具有相似的基因结构与保守基序组成;柳枝稷TIFY基因启动子区存在多种胁迫响应元件与激素信号调控元件,经定量实验证实PviTIFY10各成员在茉莉酸(jasmonic acid,JA)处理及盐与干旱胁迫下反应强烈;JAZ亚家族部分成员尤其是PviTIFY10b在花序与根的发育过程中有较高表达。根据以上结果推测TIFY基因家族可能在柳枝稷激素信号调控与胁迫防御反应及生长发育等多方面发挥功能。

Abstract: To explore biological functions of TIFY gene families in switchgrass (Panicum virgatum),this study used bioinformatic methods to identify the members of TIFY gene family in genome of switchgrass,and also analyzed the chromosomal location,evolutionary and structural characteristics,cis-acting elements in promoter region,spatiotemporal expression profile of the members. In addition,we used quantitative experiment to verify the characteristics of hormone and stress response of some members in JAZ subfamily. The results were as follows:There were a total of 48 TIFY genes in switchgrass that belonged to ZML(7),TIFY(1) and JAZ(40) subfamily. They distributed on all chromosome other than chromosome 3 and some members arranged in clusters on chromosome 9. These genes could be divided into 5 groups according to the evolutionary and structural characteristics,and the members of each group had similar gene structure and conserved motifs. A variety of regulatory elements related to stress and hormone response in promoter region of TIFY genes in switchgrass and quantitative experiments confirmed that the members of PviTIFY10 responded strongly to JA,stresses of salt and drought. Moreover,some members of the JAZ subfamily,especially PviTIFY10b,was highly expressed in the development process of inflorescence and roots. Based on the above results,it was speculated that TIFY gene family might play a role in hormone signal regulation,stress defense response and process of growth and development in switchgrass.
