桃色欧文氏菌Cp2脂多糖对紫花苜蓿幼苗生长的影响

Effect of Lipopolysaccharide in Erwinia persicina Cp2 on the growth of Medicago sativa seedlings

摘要: 为探究桃色欧文氏菌脂多糖(Erwinia persicina Cp2 lipopolysaccharides,Cp2-LPSe)对紫花苜蓿(Medicago sativa)幼苗生长的影响,本研究以紫花苜蓿'巨能551'为材料,用改良的热酚水法提取纯化Cp2-LPSe,设不同浓度(0,0.157,0.314,0.471和0.628 EU·mL-1) Cp2-LPSe处理,测定处理后种子发芽和幼苗生长指标。结果表明:改良后的热酚水法提取Cp2-LPSe的平均产率为4.27%;不同Cp2-LPSe浓度下,紫花苜蓿种子的发芽率、发芽势和发芽指数均低于对照,且发芽指数显著低于对照(P<0.05);随着Cp2-LPSe浓度的升高,幼苗的各生长指标均呈先升后降趋势,当Cp2-LPSe浓度为0.157 EU·mL-1时,各指标均达到最大值,且显著高于对照(P<0.05)。相关性和主成分分析结果表明,0.157 EU·mL-1Cp2-LPSe对紫花苜蓿幼苗生长有较大影响。综上,0.157 EU·mL-1 Cp2-LPSe对紫花苜蓿幼苗生长有促进作用,而大于此浓度紫花苜蓿生长就会受到抑制。本研究为开发紫花苜蓿促生Cp2-LPSe制剂奠定了实践和理论基础。

Abstract: To investigate the effect of Erwinia persicina Cp2 Lipopolysaccharides (Cp2-LPSe) on the growth of alfalfa seedlings,in this experiment,alfalfa 'Juneng 551' was used to extract and purify Cp2-LPSe based on the improved hot phenol water method. Alfalfa was treated with 0,0.157,0.314,0.471,and 0.628 EU·mL-1 Cp2-LPSe,and seed germination and seedling growth were investigated. The results showed that the average yield of Cp2-LPSe was 4.27%. The germination percentage,germination potential,and germination index of alfalfa seeds treated with different Cp2-LPSe concentrations were lower than those of the control,and the germination index was significantly lowered (P<0.05). With the increase of Cp2-LPSe concentration,all the growth indexes of seedlings increased at first and then decreased. With 0.157 EU·mL-1 Cp2-LPSe treated,all the germination and growth indexes reached the maximum and were significantly higher than those of the control (P<0.05). The correlation and principal component analysis confirmed that 0.157 EU·mL-1 Cp2-LPSe had a significant effect on the growth of alfalfa seedlings. In conclusion,0.157 EU·mL-1 Cp2-LPSe treatment promoted the growth of alfalfa seedlings,and was beneficial to the accumulation of biomass,but more than this concentration would inhibit the growth. This study laid a practical and theoretical foundation for the development of alfalfa growth-promoting reagent Cp2-LPSe.
