超低温保存对多年生黑麦草与红豆草种子质量的影响

Abstract: Effect of cryopreservation on seed quality of perennial ryegrass(Lolium perenne) and sainfoin(Onobrychis viciifolia Scop.) was studied in this paper by measuring and analyzing the seed germination rates,germination index,electroconductibility,dehydrogenase activity,MDA content,and soluble sugar content before and after cryopreservation at different seed moisture contents.The results show that perennial ryegrass and sainfoin seeds at natural moisture content could be cryopreserved in liquid nitrogen.There was no significant effect of cryopreservation time on the qualities of two kinds of seeds.There were adaptive changes detected for sainfoin seeds after cryopreservation vs.before cryopreservation,while no difference for perennial ryegrass.Seed moisture content was not a critical factor determining cryopreservation effects;the appropriate range of moisture content for perennial ryegrass and sainfoin seeds cryopreservation was 9.2%~11.8% and 5.6%~9.0%,respectively.

摘要: 对多年生黑麦草(Lolium perenne)及红豆草(Onobrychis viciaefoli)种子超低温保存前后种子发芽率、发芽指数、电导率、脱氢酶活性、丙二醛含量和可溶性糖含量进行测定分析,探讨超低温保存对种子质量的影响。结果表明:多年生黑麦草与红豆草种子在自然含水量状态即可直接投入液氮中保存,保存时间的长短对种子质量影响不显著;多年生黑麦草种子超低温保存前后差异不显著,红豆草种子冻后发生了有益的适应性变化;含水量不是影响超低温保存效果的重要因素,多年生黑麦草和红豆草种子超低温保存最适含水量范围分别为9.2%~11.8%与5.6%~9.0%。
