基于叶绿素荧光参数和生理指标对冰草种质资源幼苗期的耐盐性评价

Abstract: To identify salt-tolerant germplasm resources of Agropyron cristatum, this study subjected 31 germplasm samples to salt stress treatment with a soil salt content of 0.9%. Chlorophyll fluorescence parameters (phi2, NPQt, qL, Fv/Fm, Fo, Fm, Y(NPQ), Y(NO), and Y(II)) and physiological indicators (MDA, Pro, SS, and POD) were determined, and their salt tolerance was comprehensively evaluated using correlation analysis, principal component analysis (PCA), and cluster analysis. The results showed that the coefficient of variation of the measured indicators of 31 samples ranged from 0.03 to 0.99. PCA indicated that the contribution rates of the top four principal components were 43.152%, 22.847%, 9.746%, and 8.882%, respectively. Cluster analysis suggested that the first group had higher photosynthetic efficiency and POD activity, and lower MDA content; the second group had moderate photosynthetic rate; the photosynthesis and physiological indices of the third group were severely affected. Based on PCA and cluster analysis, combined with changes in physiological responses, six salt-tolerant Agropyron germplasm (20-02, 20-03, 20-06, 20-08, 20-11, 20-17) were screened, most of which belonged to A. latissimum. In conclusion, the salt-tolerant germplasm obtained in this study could be used as the candidate materials for the breeding of new varieties of Agropyron, and also provide germplasm resources for the exploration of excellent salt-tolerant genes.

摘要: 为筛选耐盐冰草(Agropyron Gaertn.)种质资源,本研究对野生采集的31份冰草种质资源开展混合中性盐胁迫处理,通过检测叶绿素荧光参数,包括φPSII,NPQt,qL,Fv/Fm,Fo,Fm,Y(NPQ),Y(NO)及Y(II)同时测定MDA,Pro,SS和POD等生理指标,运用相关性分析、主成分分析(PCA)及聚类分析,对种质耐盐性进行综合评价。结果显示,31份冰草材料测定指标变异系数介于0.03~0.99之间。主成分分析显示,前四个主成分贡献率依次为43.152%,22.847%,9.746%和8.882%。聚类分析将种质分为三类:Ⅰ类光合效率与过氧化物酶活性较高,丙二醛含量较低;Ⅱ类光合速率处于中等水平;Ⅲ类光合作用及生理指标显著受盐胁迫影响。基于主成分及聚类分析,并结合生理响应变化,筛选出6份耐盐冰草种质资源(20-02,20-03,20-06,20-08,20-11,20-17),其中多数为扁穗冰草。本研究获得的冰草耐盐种质资源可作为冰草新品种选育候选材料,也为挖掘优良耐盐基因提供种质基础。
