Abstract: Soil nematodes occupy the central link of the soil food web and have an important impact on soil ecosystems. High-throughput sequencing technology has gradually gained much attention in nematode identification due to its advantages such as high resolution,sensitivity and wide sequencing range. However,the concordance study between high-throughput sequencing technology and morphological identification results is still lack. In order to better understand the two analytical methods,this study was conducted to determine and compare the composition and structure of soil nematode communities in different patches of degraded alpine meadows in Menyuan County of the upper Yellow River region using morphological identification and high-throughput sequencing techniques. The results showed that the abundance of nematode communities had significant differences under different patches of degraded alpine meadows in Menyuan County of the upper Yellow River area and the differences were not significant in the determination of biodiversity. Although high-throughput sequencing technology provided an effective technical tool for rapid insight into nematode diversity,there were still some limitations that lead to poor accuracy of the measurement results and poor consistency with the results of morphological identifications. The improvement of high-throughput sequencing database could further improve the accuracy of ecological information obtained by molecular techniques.

摘要: 土壤线虫是土壤循环重要的环节,对土壤生态系统具有重要影响。近年来,高通量测序技术因其分辨率和灵敏度高、测序范围广等优势,在线虫鉴定方面逐渐受到关注。目前关于高通量测序技术和形态学鉴定结果的一致性研究,仍比较欠缺。为了更好地比较两种分析方法,本研究同时采用形态学鉴定和高通量测序技术,对黄河上游区门源县不同斑块化退化高寒草甸土壤线虫群落组成和结构进行了测定和比较。结果表明两种分析方法在线虫群落丰度测定上具有显著差异,但在生物多样性的趋势的判断上差异不大。尽管高通量测序技术为快速洞察线虫多样性提供了有效的技术手段,但仍存在一些局限性导致其测定结果准确性、与形态学鉴定结果的一致性较差,通过高通量测序数据库的改进,可以进一步提高分子技术获取生态信息的准确性。
