Genetic Diversity Analysis of 60 Tall Fescue Varieties Based on EST-SSR Markers

Abstract: Tall fescue (Festuca arundinacea Schred) is a perennial cool-season grass with a wide range of applications in temperate zones as a turfgrass,forage grass,and ecological restoration grass. In this study,60 tall fescue varieties from different sources were analyzed for genetic diversity using expressed sequence tag-simple sequence repeat (EST-SSR) primers,and fingerprint profiles were constructed to identify and evaluate the germplasm resources. The results showed that a total of 173 bands were amplified by 20 EST-SSR primers,163 of which were polymorphic bands,with a percentage of 94.220%;all primers identified varieties and different primer combinations can identify 60 varieties;the clustering results were correlated with geographic regions,but was not obvious;and three subgroups were determined to be the optimal number for the 60 participating materials. The results indicated that the genetic diversity of the 60 varieties was at a medium level,and the SSR markers can effectively identify tall fescue varieties and evaluate its genetic differences,laying the foundation for the construction of core tall fescue varieties and the selection and breeding of new varieties of tall fescue.

摘要: 苇状羊茅(Festuca arundinacea Schred)是多年生冷季型禾草,作为草坪草、牧草、生态修复草种在温带有着广泛的应用。本研究以60份不同来源的苇状羊茅种质为材料,利用表达序列标签-简单序列重复(EST-SSR)引物对其进行遗传多样性分析,对其构建指纹图谱,对种质资源进行鉴定与评价。结果表明:20对EST-SSR引物共扩增出173条带,多态性条带163条,多态性位点百分率94.220%;所有引物均可识别种质,不同的引物组合可以区分60份种质;聚类结果与地理区划有一定的关联,但不明显;60份参试材料最佳亚群数为3。研究结果表明:60份种质的遗传多样性处于中等水平;SSR标记可以有效地鉴定苇状羊茅种质并评价其遗传差异,为苇状羊茅核心种质的构建、新品种的选育奠定基础。
