西藏野生垂穗披碱草CnCOR410基因克隆及表达分析

Abstract: To investigate the molecular mechanism of dehydrin regulating plant abiotic stress response, the CnCOR410 gene was cloned from Xizang wild Campeiostachys nutans Griseb. Amino acid sequence analysis, subcellular localization, and expression profile of the CnCOR410 gene in response to cold, drought, and salt stress were conducted. The results showed that the coding sequences (CDS) oftheCnCOR410 gene was 765 bp in length, encoding 254 amino acids, with a molecular weight of 7504.48 Da and an isoelectric point of 4.87. The CnCOR410 protein was a hydrophilic and non-transmembrane protein, and its secondary structure was mainly random coil, accounting for 61.42%. The CnCOR410 contained the conserved domain of dehydrin and belonged to the LEA family. The amino acid sequence similarity between CnCOR410 and wheat (Triticum aestivum L) was the highest at 93.44%, and the protein was located in the cytoplasm. The RT-qPCR results showed that CnCOR410 was expressed in the root, stem, and leaf tissues of C. nutans, with the highest expression in the leaves. Moreover, the expression of CnCOR410 was regulated by cold, drought, salt stress, and ABA (P<0.05). The results of this study laid a foundation for further clarifying the mechanism of CnCOR410 regulating the abiotic stress adaptation of C. nutans.

摘要: 为了研究脱水素调控植物非生物胁迫应答的分子机制,本研究从西藏野生垂穗披碱草(Campeiostachys nutans Griseb.)中克隆了CnCOR410基因,并进行了生物信息学、基因表达模式和亚细胞定位分析。结果表明:CnCOR410序列全长765 bp,编码254个氨基酸,分子量为7504.48 Da,等电点为4.87;CnCOR410蛋白属于亲水蛋白、非跨膜蛋白;二级结构以无规则卷曲为主,占比61.42%;CnCOR410包含脱水素保守结构域属于LEA家族;CnCOR410与小麦(Triticum aestivum L.)的氨基酸序列相似性最高为93.44%,且该蛋白定位在细胞质上;CnCOR410在垂穗披碱草根、茎、叶组织中均有表达,在叶片中表达量最高;且CnCOR410表达受低温、干旱、盐胁迫和ABA调控(P<0.05)。本研究结果为深入探明CnCOR410调控垂穗披碱草非生物胁迫适应机制奠定了基础。
