外源草酸对不同年限狗尾草种子萌发及生理生化的影响

Effect of Exogenous Oxalic Acid on Germination and Physiological Biochemical Characteristics of Setaria sphacelata ‘Narok’ Seeds of Different Years

摘要: 以‘纳罗克’狗尾草(Setaria sphacelata ‘Narok’)种子为供试材料,采用不同浓度(5~25 mmol·L-1)和不同时间(5~25 min)的草酸处理,探究外源草酸对非洲狗尾草种子萌发(发芽率、发芽势、发芽指数和活力指数)及生理特征(可溶性糖含量、超氧化物歧化酶(Superoxide dismutase,SOD),过氧化物酶(Peroxidase,POD),过氧化氢酶(Catalase,CAT)活性、丙二醛(Malondialdehyde,MDA)含量)的影响,并研究了外源草酸对储藏0~4年狗尾草种子的生理特性及幼苗生长的影响。结果表明:草酸处理对收获当年狗尾草种子的萌发具有促进作用,随着处理浓度和时间增加至25 mmol·L-1,25 min,收获当年的种子可达到破眠效果;采用25 mmol·L-1,25 min的条件处理不同储藏年限的种子,显著提高收获当年种子的SOD和CAT活性,增加0~2年种子幼苗的根长,降低种子MDA含量,POD活性随储藏年份的延长逐渐降低。草酸处理后不同储藏年份种子的综合评分为0年>1年>2年>3年>4年。推荐采用外源草酸处理条件为:25 mmol·L-1,25 min,可有效促进收获当年和储藏1年种子的萌发及生长。

Abstract: Hydroponic experiment was conducted to understand the effects of different concentrations of exogenous oxalic acid (5, 10, 15, 20, 25 mmol·L-1) and different times (5, 15, 25 min) on germination (germination rate, germination energy, germination index and vitality index), physiological characteristics (soluble sugar content, SOD, POD, CAT enzyme activity and MDA content) and seedling growth of Setaria sphacelata ‘Narok’ seeds stored for 0, 1, 2, 3 and 4 years. The results showed that germination energy, germination index and vitality index of seeds harvested at the first the year increased significantly with increasing oxalic acid concentrations and treated time. 25 mmol·L-1oxalic acid treatment for 25 min could break seed dormancy with 84.0% seed germination rate and 16.3 vitality index, respectively, which were 10.1 and 12.5 times higher than those in CK. The root length of seedlings from 0 to 2 a of storage seed effectively increased under 25 mmol·L-1 oxalic acid for 25 min treatment, and the root length of seedlings from 2 a of storage seed increased by 226.6% compared with the CK. POD activities gradually decreased with increasing the years of storage. POD activities of seeds in the year of harvest decreased by 22.2% compared with CK. The SOD and CAT activities of seeds in the year of harvest increased by 87.3% and 164.4%, respectively. The MDA contents of seeds stored for 0~2 a effectively decreased. MDA contents of seeds in the year of harvest reduced by 35.4%. The overall scores of seeds in different storage years after oxalic acid treatment were 0 a > 1 a > 2 a > 3 a > 4 a. Therefore, 25 mmol·L-1exogenous oxalic acid for 25 min treatment could effectively promote the germination and growth of seeds in the year of harvest and the one year of storage.
