红花绿绒蒿种子休眠解除的生理生化响应

Physiological and Biochemical Responses of Meconopsis punicea Maxim. to Dormancy Release

摘要: 为了探究红花绿绒蒿(Meconopsis punicea Maxim.)种子休眠解除对胚发育及生理生化变化的响应,本研究对红花绿绒蒿种子进行变温层积处理,观测不同层积处理下种子的胚形态、胚率、发芽率,测定种子营养物质及内源激素含量、呼吸途径关键酶活性等理化指标。结果表明:层积后,红花绿绒蒿种胚完成形态后熟,发芽率显著提高(P<0.05);淀粉、可溶性糖、可溶性蛋白含量显著升高(P<0.05);丙酮酸激酶(Pyruvate kinase,PK)活性显著降低(P<0.05),琥珀酸脱氢酶(Succinate dehydrogenase,SDH)活性、6-磷酸-葡萄糖脱氢酶(Glucose-6-phosphate dehydrogenase,G-6-PDH)活性显著升高(P<0.05);赤霉素(Gibberellic acid,GA)含量无显著变化,脱落酸(Abscisic acid,ABA)含量、吲哚乙酸(Indole-3-acetic acid,IAA)含量及ABA/GA3,ABA/GA1比值显著降低(P<0.05)。综上所述,低温层积能够解除红花绿绒蒿种子休眠,种子内营养物质积累,磷酸戊糖途径(Pentose phosphate pathway,PPP)途径活化,ABA,IAA含量及ABA/GA3,ABA/GA1比值下降是解除红花绿绒蒿种子休眠的关键。

Abstract: In order to explore the response of the seeds of Meconopsis punicea Maxim. to dormancy release and its embryo development,physiological and biochemical changes after that release,the seeds of Meconopsis punicea were subjected to the stratification treatments with various temperatures,and the embryo morphology,embryo rate,and germination rate of the seeds in different stratification treatments were observed;and the content of nutrients and endogenous hormones in the seeds,as well as the activity of key enzymes in the respiratory pathway and other physicochemical indicators were also measured. The results showed that:after stratification treatments,the embryo of M. punicea completed its morphological ripening and the germination rate significantly increased (P<0.05);the content of starch,soluble sugar and soluble protein all significantly increased (P<0.05);the PK activity significantly decreased (P<0.05),while succinate dehydrogenase (SDH) and glucose-6-phosphate dehydrogenase (G-6-PDH) activities significantly increased (P<0.05). There was no significant change in the content of GA3 and GA1,while the content of ABA and IAA,as well as the ratios of ABA to GA3 and ABA to GA1 significantly decreased (P<0.05). In conclusion,the low temperature stratification can relieve the seed dormancy of M. punicea,the accumulation of nutrients in seeds,activation of PPP pathway,reduction of ABA,IAA contents and ratios of ABA to GA3 and ABA to GA1 are the key factors to release the seed dormancy in M. punicea.
