柳枝稷GRF转录因子家族全基因组鉴定与分析

摘要: 生长调节因子(Growth-regulating factor,GRF)家族在植物生命进程中发挥关键作用。本研究共鉴定了19个柳枝稷(Panicum virgatum L.)PvGRF基因,它们不均匀的分布在11条染色体上,可分为4个亚家族,同一亚家族成员具有相似的模体特征和基因结构。柳枝稷PvGRF基因的扩张来自于节段性复制事件;其中15个柳枝稷基因和11个水稻基因被鉴定为直系同源基因。PvGRF基因在花和种子等生殖器官中强烈表达,这也是该家族成员调控植物生长发育的证据。PvGRF基因主要包含7种胁迫和10种激素相关的顺式作用元件;同时qRT-PCR分析表明柳枝稷GRF基因在NaCl、聚乙二醇、脱落酸、茉莉酸甲酯和水杨酸处理下差异表达,暗示着PvGRF基因在响应胁迫和激素反应中发挥作用。综上所述,本研究系统分析了柳枝稷PvGRF家族的特征及其在胁迫和激素下的表达模式,为柳枝稷GRF基因的功能研究提供基础。

Abstract: Growth-regulating factor (GRF) family plays a key role in plant life progress. In the present study,19 switchgrass (Panicum virgatum L.) PvGRF genes were identified and distributed unevenly on 11 chromosomes and were clustered into four subfamilies. PvGRF members belonging to the same subfamily shared similar motif composition and gene structure. Synteny analysis revealed that segmental duplications played significant roles in switchgrass GRF gene expansion,and 15 switchgrass genes and 11 rice genes were identified as homologous genes. The PvGRF genes were strongly expressed in reproductive tissues such as flowers and seeds,indicating that PvGRFs play critical roles in plant growth and development. The PvGRF genes mainly contain 7 stress and 10 hormone-related cis-acting elements. Meanwhile,qRT-PCR analysis showed that the switchgrass GRF genes were differentially expressed under NaCl,PEG,ABA,MeJA,and SA treatments,which could prove that PvGRF members play vital roles in response to stress and hormones treatments. In summary,the study systematically analyzed the characterization of the switchgrass PvGRF family and its expression patterns under stress and hormones,and provided a basis for the functional research of switchgrass PvGRF genes.
