Abstract: The Bcl-2-associated athanogene (BAG) protein family is a multifunctional family that plays a critical role in plant growth and stress responses. However, the BAG gene family has not yet been systematically characterized in alfalfa (Medicago sativa). In this study, we analyzed the physicochemical properties, evolutionary relationships, gene structures, conserved motifs, cis-regulatory elements, chromosomal localization, collinearity, and gene expression patterns of the BAG gene family in alfalfa using bioinformatics methods and transcriptome data. The results revealed that the alfalfa BAG gene family consists of eight members, which are unevenly distributed across five chromosomes. Subcellular localization predictions indicated their presence in the nucleus, chloroplasts, and/or cytoplasm. Phylogenetic analysis classified the alfalfa BAG genes into three subfamilies. Cis-regulatory element analysis showed that the alfalfa BAG gene family contains numerous elements associated with plant growth and development, light responsiveness, hormone responses, and biotic/abiotic stress responses. The MsBAG gene family exhibited significant responses to abiotic stresses such as abscisic acid (ABA), low temperature, drought, and salinity. This study provides a fundamental theoretical basis for further investigation into the potential roles of the BAG gene family in abiotic stress tolerance in alfalfa.
