偏关苜蓿种子萌发及植株再生体系优化

Seed Germination and Plant Regeneration System Optimization of Medicago sativa ‘Pianguan’

摘要: 本试验以偏关苜蓿(Medicago sativa ‘Pianguan’)种子为材料,从不同消毒剂、培养基成分对种子萌发以及不同激素及浓度配比对愈伤组织诱导分化的影响等方面展开研究。结果表明:0.1% HgCl2消毒6 min后置于1/4MS培养基上的种子发芽率、发芽势较高,霉烂率低;不同外植体的最优愈伤诱导培养基为MS+2 mg·L-1 2,4-D+0.4 mg·L-1KT,下胚轴与子叶最高诱导率分别为100%和98%;最优愈伤分化培养基为UM+1.6 mg·L-1KT,下胚轴与子叶最高分化率分别为59%和44%;不定芽转至1/2MS生根培养基,生根率达64%。本研究系统构建了优化的偏关苜蓿植株再生体系,可为后续遗传转化提供技术支撑。

Abstract: In this experiment,the seed germination characteristics and tissue culture methods of Medicago sativa ‘Pianguan’ were studied by selecting the optimized disinfectant and medium components on seed germination,and concentration ratio of different hormones on callus induction & differentiation. The results showed that the Medicago sativa ‘Pianguan’ showed high germination percentage,high germination potential and low mildew percentage when the seeds were disinfected with 0.1%HgCl2for 6 min,and then inoculated on 1/4 MS medium. The optimal callus induction medium for different explants was that with MS+2 mg·L-12,4-D+0.4 mg·L-1KT,the highest induction percentage of hypocotyls and cotyledons were 100% and 98% respectively. The UM+1.6 mg·L-1KT was the best medium for callus differentiation,the highest differentiation percentage of hypocotyls and cotyledons were 59% and 44% respectively. When adventitious shoots were transferred to 1/2 MS rooting medium,the rooting percentage was 64%. The work built a complete and optimized regeneration system of Medicago sativa ‘Pianguan’,which can provide technical support for the subsequent genetic transformation.
