Effects of Different Culture Media on the Leaf Regeneration of Chicory

摘要: 以3个菊苣(Cichorium intybus L.)品种叶片为外置体,以MS,N6,B5,SH和N6B5MS为基本培养基,添加不同激素对其不定芽诱导及植株再生进行研究。结果表明:3个菊苣品种在5种培养基上的愈伤诱导率和分化率存在差异,其中以MS培养基添加2.0 mg·L-16-BA和0.2 mg·L-1 IBA再生和分化最好,平均愈伤诱导率和分化率分别为70.12%和72.07%。在不同生根培养基中,生根率和根长存在一定的差异,生根数量没有显著差异,其中以1/2MS培养基中添加0.1 mg·L-1的NAA效果最显著,生根率、每个外植体的平均根数和根长分别达到97.07%,4.03个和6.51 cm。

Abstract: Efficient adventitious shoot induction and plant regeneration of young leaves from three chicory cultivars on MS, N6, B5, SH and N6B5MS media with different hormone concentrations were studied in this paper. Results showed that average callus induction rate and differentiation rate had significant difference among three chicory cultivars on five media. The best medium was MS supplemented 2.0 mg·L-1 6-BA and 0.2 mg·L-1 IBA, average callus induction rate and differentiation rate were 70.12% and 72.07%, respectively. Rooting rate and length had significant difference between MS and 1/2MS media, whereas the number of root had no significant difference. The best root medium was 1/2MS supplemented 0.1 mg·L-1NAA, Rooting rate, the length and number of root were 97.07%, 6.51 cm and 4.03, respectively.
