耐低温乳酸菌的筛选鉴定及其对黑麦草青贮的影响

Identification of Lactic Acid Bacteria Isolates and Their Inoculating Effects on the Silage Fermentation of Italian Ryegrass at Low Temperature

摘要:

本研究通过限制性培养方法,以温度和pH值为限制因素筛选耐低温菌株;并利用生理生化及16S rDNA同源序列分析方法综合鉴定筛选菌;将筛选菌分别添加于多花黑麦草(Lolium multiflorum)新鲜和晾晒青贮原料中,观察其在低温条件下的青贮发酵效果。结果表明:LT1,LT2及LT3均为植物乳杆菌(Lactobacillus plantarum),可在10℃,pH4.0下生长。单纯晾晒可提高黑麦草的青贮品质。15℃条件下,无论是否晾晒,添加3株耐低温乳酸菌均可促进乳酸发酵,降低pH值,减少蛋白质或氨基酸的分解,显著提高青贮饲料的发酵品质,效果优于对照菌,而2个对照菌对晾晒的黑麦草无效果。

Abstract:

Psychrotolerant strains of lactic acid bacteria were screened through continuous restricted cultivation, using pH and temperature as the indicators. Selected strains were identified via morphological, physiological, biochemical and 16S rDNA homology sequence analysis. At the same time, the effects of selected strains on the fermentation quality of fresh and wilted Italian ryegrass at low temperature were investigated. The results showed that all three strains (LT1, LT2 and LT3) were Lactobacillus plantarum, and could grow at 10℃ and pH 4.0. Wilting alone improved the fermentation quality of Italian ryegrass ensiled at 15℃. LT1, LT2 and LT3 were most effective at improving the fermentation quality of Italian ryegrass silage, irrespective of wilting, as indicated by lower pH values, less NH3-N and more lactic acid. The commercial inoculant LQ (L. plantarum and P. acidilactici) and L. rhamnosus HT1 benefited the silage fermentation of un-wilted ryegrass material, but did not affect the wilted material.
