芥子油苷对豌豆蚜生长、存活及保护酶的影响

Effects of Glucosinolates on the Growth,Survival and Protective Enzymes of Pea Aphid (Acyrthosiphonpisum)

摘要: 为了明确芥子油苷对2种色型豌豆蚜生长、存活及体内保护酶的影响。本研究采用豌豆蚜(Acyrthosiphon pisum)全纯人工饲料薄膜饲养技术,通过添加不同浓度(CK∶0 μmol·g-1;A1∶0.94 μmol·g-1;A2∶1.87 μmol·g-1;A3∶2.86 μmol·g-1)的芥子油苷,测定了豌豆蚜的存活率、相对日均体重增长率及保护酶活性等指标。结果表明:2种色型豌豆蚜的存活率和相对日均体重增长率下降;有翅蚜率逐渐升高,在2.86 μmol·g-1浓度时达到最大,红色型为46.0%、绿色型为43.3%;同时,豌豆蚜体内的超氧化物歧化酶(Superoxide dismutase,SOD)、过氧化物酶(Peroxisome,POD)和过氧化氢酶(Hydrogen peroxidase,CAT)酶活性升高,而可溶性蛋白总量降低(P<0.05);此外,相对于取食正常人工饲料的豌豆蚜,取食含有芥子油苷的红色型豌豆蚜体内SOD和CAT的响应程度明显高于POD,绿色型豌豆蚜体内POD的响应程度明显高于SOD和CAT。研究结果表明,不同色型豌豆蚜体内的保护酶系统对芥子油苷的响应机制有所不同。

Abstract: To clarify the effects of glucosinolates on the growth, survival and protective enzymes of two colored pea aphid(Acyrthosiphon pisum), the survival rate, the growth rate of relative average daily body weight and protective enzyme activity of pea aphids were determined by adding different concentrations of glucosinolates (CK:0 μmol·g-1, A1:0.94 μmol·g-1, A2:1.87 μmol·g-1, A3:2.86 μmol·g-1) using a pea aphid full-pure artificial diet film feeding technology. The results showed that the glucosinolates had a certain inhibitory effect on the growth and survival with two colored pea aphids. The rate of winged aphid, which was 46.0% in red aphid and 43.3% in green was increased gradually and reached the maximum at 2.86 mol/g. Meanwhile, the activities of Superoxide Dismutase(SOD), Peroxisome(POD) and Hydrogen Peroxidase(CAT) in aphids were significantly increased, however, the total amount of soluble protein was decreased. Moreover, compared with normal feeding pea aphid, the response level of SOD and CAT in red type pea aphid feeding with glucosinolin was significantly higher than that of POD, and the response level of POD in green was significantly higher than that of SOD and CAT. The results showed that the protective enzyme system in pea aphid to glucosinolates with different color types had different response mechanisms.
