苜蓿褐斑病的离体叶接种研究

Study on Excised Leaf Tissue Inoculation of Common Leaf Spot in Alfalfa

摘要: 研究和鉴定苜蓿褐斑病抗病性的离体叶接种技术。结果表明,离体叶接种鉴定比传统的整株鉴定效率高,结果可靠。在0.3%水琼脂培养基中加入50μm/mL苯骈咪唑,即可达到最佳保绿效果。接种所需的适宜条件是,相对湿度接近100%,温度20℃,接种时间20h,无光照。培养所需的适宜条件是,生长箱内光照(光强度9000lux)和黑暗每12h交替一次,有光时22℃,无光时18℃。

Abstract: The excised leaf tissue inoculation (ELTI) method is studied in this paper for identification of alfalfa resistance to common leaf spot. The results showed that ELTI is more efficient and dependable in resistance identification than the traditional whole plant inoculation method. Greenness of excised leaf tissues was the best maintained by adding benzimidazole into media of 0.3% water agar at 50μm/mL. The conditions required for a good inoculation of excised leaf tissue includes relative humidity about 100%, temperature of 20℃, duration of 20 hours, and complete dark. The condition required for good culture are a 12 hours interval alternation of light (9000lux) with temperature at 22℃ and dark with temperature at 18℃.
