不同品种紫花苜蓿种子萌发期耐低温性综合评价

摘要: 低温是限制植物正常生长发育的常见胁迫因子,尤其对种子萌发过程造成严重影响。本研究以22个品种紫花苜蓿(Medicago sativa L.)种子为试验材料,通过测定标准发芽温度(20℃,即对照)和低温(10℃)两种条件下9个种子萌发和幼苗生长特性相关指标变化,运用耐冷系数、隶属函数、聚类分析方法对种子萌发期耐低温性进行综合评价,以期为紫花苜蓿耐低温种质筛选与新品种培育研究提供理论参考。结果表明:低温胁迫下,各品种紫花苜蓿的9个种子萌发和幼苗生长特性相关指标均呈现下降趋势。基于综合耐冷系数与隶属函数进行系统聚类分析,将22个品种紫花苜蓿划分为4个不同耐低温类型,即强耐低温性、中耐低温性、弱耐低温性和低温敏感性,并筛选出强耐低温性品种为‘赛迪10号’、低温敏感性品种为‘英斯特’,为紫花苜蓿耐低温种质筛选和种植利用提供了基础数据,也为其耐低温基因挖掘及新种质创制奠定了理论基础。

Abstract: Low temperature is a common stress factor that limits the normal growth and development of plants,especially impose serious impacts on the seed germination process. In this study,22 varieties of alfalfa (Medicago sativa L.) seeds were used as experimental materials to measure the changes of nine indicators of seed germination and seedling growth characteristics under two temperature conditions,that is the standard temperature (20℃,i.e. control) and the low temperature (10℃). The cold tolerance coefficient,membership function,and cluster analysis method were used to comprehensively evaluate the low temperature tolerance of alfalfa during seed germination,which would provide theoretical reference for the screening of low temperature tolerant germplasm and breeding of new varieties. The results indicated that all the nine indicators related to seed germination and seedling growth characteristics of 22 alfalfa varieties showed the downward trend under low temperature stress. Based on the comprehensive cold tolerance coefficient and membership function for systematic clustering analysis,22 varieties of alfalfa were divided into four different cold tolerance types,namely strong low temperature tolerance,medium low temperature tolerance,weak low temperature tolerance,and low temperature sensitivity. Then,the ‘Sardi 10’ was selected as a variety with strong low temperature tolerance,and the ‘Instict’ as a variety with low temperature sensitivity,respectively. This study provided some basic data for the screening and utilization of alfalfa germplasm with low temperature tolerance,and also laid a theoretical foundation for its gene mining associated with low temperature tolerance and new germplasm creation.
