辐射白刺花遗传多样性的ISSR和RAPD比较分析

Comparison of Genetic Diversity of Irradiated Sophora viciifolia Using ISSR and RAPD Markers

摘要:

采用ISSR和RAPD标记对8个60Co-γ辐射梯度诱变白刺花(Sophora viciifolia)进行遗传多样性分析。结果表明:20条ISSR和6条RAPD引物的PPL多态率分别为51.37%和21.03%,ISSR的Nei's指数和平均Shannon指数均大于RAPD;ISSR的标记指数(1.28)是RAPD(0.20)的6.4倍,表明ISSR具有较高的标记效率。Mantel检测表明,ISSR+RAPD标记与ISSR标记的结果显著相关(P<0.05)。综合分析表明白刺花用ISSR标记可检测出更高的多态性,更能准确反映白刺花材料间的遗传关系。

Abstract:

The RAPA and ISSR markers were used in analyzing the genetic diversity of mutagenic Sophora viciifolia Franch with eight different 60Co-γ-rays gradients. The results showed that the percentage of polymorphic bands of 20 ISSR primers and 6 RAPD primers were 51.27% and 21.03%, respectively. The Nei's gene heterozygosity and Shannon's information index of ISSR were greater than RAPD. For 8 materials, ISSRs got a MI value(1.28) that was 6.4 times higher than that of RAPDs(0.20), indicating that ISSRs had a higher MI efficiency. The Mantel test revealed that the correlation was significant between ISSR+RAPDs and ISSRs(r=0.9791, P<0.05). Comprehensive research showed that ISSRs could detect the higher polymorphism of Sophora viciifolia, and more accurately reflect the genetic relationship among Sophora viciifolia.
