The Effects of Different Chemical Reagents on Germination Characteristics of Allium senescens L. Seed

摘要: 为探明多菌灵(Carbendazim,MBC)、过氧化氢(Hydrogen peroxide,H2O2)、高锰酸钾(Potassium permanganate,KMnO4)对山韭(Allium senescens L.)种子萌发过程的影响,本研究以山韭(Allium Senescens L.)种子为试验材料,分析不同浓度MBC(100,200,300,400,500 倍)、H2O2(0.5%,1.0%,1.5%,2.0%,2.5%)、KMnO4(0.1%,0.2%,0.4%,0.8%,1.6%)处理下种子萌发指标、生理指标及内源激素的变化规律。结果表明:与CK相比,200倍MBC、1.0% H2O2、0.4% KMnO4对山韭种子的萌发有显著促进作用(P<0.05),可溶性糖含量、脂肪酶活性、中性蛋白酶活性、乙烯(Ethrel,ETH)含量均显著提高(P<0.05)。主成分分析和相关性分析表明,MBC和KMnO4处理的山韭种子发芽率、发芽势、发芽指数和活力指数与酶活性呈正相关关系;H2O2处理的山韭种子发芽率、发芽势、发芽指数和活力指数与可溶性糖、可溶性蛋白有正相关关系。综合分析表明,200倍MBC、1.0% H2O2主要通过显著提高山韭胚芽内源ETH含量(P<0.05),而0.4% KMnO4通过显著提高内源ETH含量和内源赤霉素(Gibberellins,GA3)含量(P<0.05)来促进山韭种子萌发。

Abstract: The objective of this research was to study the effects of Carbendazim (MBC),Hydrogen peroxide (H2O2) and Potassium permanganate (KMnO4) on the germination of Allium senescens L. seeds. To conduct the experiments,A. senescens seeds were used and subjected to various concentrations of MBC (100,200,300,400,500-fold dilution),H2O2 (0.5%,1.0%,1.5%,2.0%,2.5%) and KMnO4 (0.1%,0.2%,0.4%,0.8%,1.6%). The changes of germination indexes,physiological indexes and contents of endogenous hormones in different exogenous additives were analyzed. The results showed that compared with CK,200 times MBC,1.0%H2O2,0.4%KMnO4 significantly promoted seed germination(P<0.05),and significantly increased soluble sugar content,lipase enzymatic activity,neutral protease enzymatic activity and Ethrel (ETH) content (P<0.05). The results of principal component analysis and correlation analysis showed that the germination potential,percentage of germination,germination index and vital index of the seeds treated with MBC and KMnO4 had a positive correlation with the enzyme activity. The germination energy,percentage of germination,germination index and vital index of the seeds treated with H2O2 were linked to soluble sugar and soluble protein. The comprehensive analysis showed that 200 times MBC and 1.0% H2O2increased the endogenous ETH content of A. senescens plumule to improve the germination ability of A. senescens seeds(P<0.05),and 0.4% KMnO4 not only increased the endogenous ETH content of A. senescens (P<0.05),but also increased the endogenous Gibberellins (GA3) content to promote the seed germination(P<0.05).
