抗坏血酸引发对燕麦老化种子胚细胞抗氧化性能的影响

Effect of AsA Priming on the Oxidation Resistance of Embryonic Cells of Aged Oat Seeds

摘要: 本研究探讨了外源抗坏血酸(Ascorbic acid,AsA)引发对燕麦(Avena sativa)老化种子胚细胞抗氧化性能的影响,以期为贮藏种子的科学利用提供理论依据。试验以老化20 d的燕麦种子为材料,用浓度为0.5,1.5和2.5 mmol·L-1的外源AsA引发0(CK),4,8,16和24 h后,分析其种胚细胞抗氧化酶活性及脂质过氧化水平的变化规律。结果表明:外源AsA引发下燕麦老化种子胚细胞的丙二醛含量显著低于CK(P<0.05),而其各抗氧化酶的活性显著高于CK(P<0.05)。低浓度(0.5 mmol·L-1)外源AsA引发24 h时,燕麦老化种子胚细胞内各抗氧化酶活性显著高于其他引发时间(P<0.05),但高浓度(1.5和2.5 mmol·L-1)外源AsA引发24 h时,燕麦老化种子胚细胞内抗氧化酶活性均显著下降(P<0.05),且其丙二醛含量上升。综上所述,外源AsA引发对燕麦老化种子胚细胞的抗氧化性能的影响与其浓度和引发时间密切相关,0.5 mmol·L-1的AsA引发24 h是提高燕麦老化种子胚细胞抗氧化性能的最佳处理。

Abstract: To provide a theoretical basis for the scientific utilization of stored seeds,the study examined the influence of exogenous ascorbic acid (AsA) priming on the antioxidant properties in embryonic cells of aged oat (Avena sativa) seeds. Oat seeds were aged for 20 d as the experimental materials,and then primed with 0.5,1.5 and 2.5 mmol·L-1 AsA for 0 (the control),4,8,16 and 24 h,and the activities of antioxidant enzymes and lipid peroxidation in their embryonic cells were tested. The results showed that the malondialdehyde contents of the embryonic cells of aged oat seeds undergone AsA priming were significantly lower than that of CK (P<0.05),but the activities of antioxidant enzymes were significantly higher than that of CK (P<0.05). Low concentration (0.5 mmol·L-1) AsA primed for 24 h,the activities of antioxidant enzymes in embryonic cells of aged oat seeds were significantly higher than those of other priming times. However,the activities of antioxidant enzymes decreased significantly in embryonic cells of aged oat seeds undergone a high concentration (1.5 and 2.5 mmol·L-1) AsA primed for 24 h (P<0.05),and their malondialdehyde contents were also increased. In conclusion,the influence of exogenous AsA priming on the antioxidant properties of embryonic cells of aged oat seeds was closely related to the AsA concentration and priming time,and priming with 0.5 mmol·L-1 AsA for 24 was the best treatment to improve the antioxidant properties of embryonic cells of aged oat seeds.
