黑果枸杞GATA转录因子家族鉴定与生物信息学分析

Identification and Bioinformatics Analysis of GATA Transcription Factor Family in Lycium ruthenicum

摘要: 为研究黑果枸杞(Lycium ruthenicum)GATA转录因子家族在盐逆境胁迫下的作用,本文对不同浓度NaCl盐胁迫下的黑果枸杞进行转录组测序和分析,筛选GATA转录因子家族成员,并分析其理化性质、保守结构域、系统发育树及表达模式。结果表明,黑果枸杞中共包含34个GATA成员,分为4个亚族,其氨基酸数目介于94~540 aa之间,分子量在10 223.79~60 326.46 Da之间,等电点介于5.64~10.22之间。保守基序的分析表明,LrGATAs成员多含C-X2-C-X17-20-C-X2-C锌指结构域,motif 1是LrGATAs较为保守的基序。盐胁迫下不同LrGATAs基因的表达模式存在差异。荧光定量PCR结果表明,低盐胁迫后,根中LrGATA6LrGATA13和叶中LrGATA16LrGATA23基因表达量上调,高盐胁迫后,根中LrGATA23,叶中LrGATA19LrGATA23基因表达量上调。本研究结果可为深入研究黑果枸杞GATA家族基因的功能及其在盐胁迫抗性中的应用提供科学依据。

Abstract: To investigate the role of GATA transcription factor family in Lycium ruthenicum under stress conditions, this study conducted transcriptome sequencing and analysis on Lycium ruthenicum under different NaCl salt stress concentrations, screened GATA transcription factor family members, and analyzed their physical and chemical properties, conserved structural domains, phylogenetic trees, and expression patterns. The results showed that Lycium ruthenicum contains 34 GATA members, divided into four subfamilies, with amino acid numbers ranging from 94 to 540 aa, molecular weights ranging from 10 223.79 to 60 326.46 Da, and isoelectric points ranging from 5.64 to 10.22. The analysis of conserved motifs showed that LrGATAs members often contain a C-X2-C-X17-20-C-X2-C zinc finger structural domain, motif 1 is a relatively conserved motif in LrGATAs. The expression patterns of different LrGATAs genes under salt stress were different. The results of qPCR showed that after low salt stress, the expression of LrGATA6LrGATA13 in roots and LrGATA16LrGATA23 in leaves was upregulated, while after high salt stress, the expression of LrGATA23 in roots and LrGATA19LrGATA23 in leaves was upregulated. The results of this study can provide scientific evidence for further research on the functions of GATA family genes in Lycium ruthenicum and their application in salt stress resistance.
