黑果枸杞XTH转录因子家族鉴定与生物信息学分析

Identification and Bioinformatics Analysis of XTH Transcription Factor Family in Lycium ruthenicum

摘要: 为探究木葡聚糖内糖基转移酶/水解酶(Xyloglucan endotransglucosylase/hydrolase,XTHs)基因家族在黑果枸杞(Lycium ruthenicum)生长发育中的作用,以及对盐胁迫的响应,本文通过生物信息学方法对XTHs基因的理化性质、保守结构域、进化关系和表达模式进行分析。从黑果枸杞转录组数据中共筛选出19个LrXTHs成员,其编码氨基酸的数目为258~330 aa,分子量为29.77~37.98 kDa,等电点为4.98~8.92,基因长度为954~1 814 bp。LrXTHs成员都含DE(I/L/F/V)D(F/I/M)EFLG保守结构域。系统发育树显示XTH基因家族可分为4个亚族。基因表达谱显示不同组织中的LrXTHs在盐胁迫下具有不同的表达趋势,LrXTH3LrXTH18LrXTH19在根部组织中高表达,LrXTH9在根部和叶部组织中高表达,表明LrXTHs可能在黑果枸杞的盐胁迫响应中发挥重要作用。该结果为黑果枸杞及XTH家族的进一步研究提供了参考。

Abstract: In order to explore the role of xyloglucan endoglycosyltransferase/hydrolase gene family (XTHs) in the growth and development of Lycium ruthenicum and their response to salt stress,the physicochemical properties,conserved domains,evolutionary relationships and expression patterns of XTHs were analyzed by bioinformatics methods. 19 LrXTHs members were screened out from the transcriptome of L. ruthenicum,with 258~330 amino acids,molecular weights of 29.77~37.98 kDa,isoelectric points of 4.98~8.92,and gene lengths of 954~1 814 bp. All members of LrXTHs contain DE(I/L/F/V)D(F/I/M)EFLG conserved domain. The Evolutionary tree showed that the XTH gene family could be divided into 4 subfamilies. Gene expression profiles showed that LrXTHs expressed in different tissues had different express trends under salt stress,the LrXTH3,LrXTH9,LrXTH18 and LrXTH19 were highly expressed in root tissues,while LrXTH9 was highly expressed in root and leaf tissues. The high expression of those LrXTH genes in root tissues of L. ruthenicum indicates that LrXTHs may play an important role in the salt stress response of the species. The results will provide a reference for the further study of L. ruthenicum and XTH family.
