SRAP标记对8份假俭草材料的鉴定分析

Identification Analysis of Eight Centipedegrass Materials by SRAP Molecular Markers

摘要: 应用SRAP分子标记技术对6个假俭草(Eremochloa ophiuroides)优良品系和2个引进品种进行扩增扫描,从分子的角度对8份材料进行鉴定。结果表明:31对SRAP引物在8份材料中扩增出576条清晰的谱条,其中多态性条带359个,多态性比率为62.33%,8份材料间的遗传相似系数变异范围为0.6962~0.8177,平均遗传相似系数为0.7388。31对SRAP引物中有1对引物Me18Em17可以将所有材料区分开来,应用该引物的扩增谱带构建了参试材料的DNA指纹图谱,可将不同材料从DNA水平上加以区分。

Abstract: Six excellent strains and two cultivars of centipederass were amplified by 31 SRAP primers. As a result, a total of 576 clear bands were detected and 359 were polymorphic with PPB (Percentage of Polymorphic Bands) at 62.33%. The genetic similarity among eight tested materials ranged from 0.6962 to 0.8177 with the average of 0.7388.One primer of Me18Em17 was selected to construct DNA fingerprint of 8 tested materials. It was able to distinguish each tested cultivar from others by polymorphic bands.
